TB-500

TB-500 is a synthetic peptide associated with tissue-repair research, usually referring to an acetylated seven-amino-acid fragment of thymosin beta-4, a naturally occurring 43-amino-acid peptide. The distinction matters because TB-500 and full-length thymosin beta-4 are different molecules, so results obtained with the full peptide do not automatically establish what the fragment does in humans. Thymosin beta-4 binds actin, a protein that forms the internal scaffolding cells reorganize to change shape, move, and participate in wound repair. TB-500 contains the sequence LKKTETQ, corresponding to an actin-binding region of thymosin beta-4 that has been investigated for repair-related activity. Experiments with this region have demonstrated promotion of angiogenesis, the formation of new blood vessels that can supply injured tissue with oxygen and nutrients.

Animal research has also found that thymosin beta-4 and a synthetic peptide containing its actin-binding sequence improve wound repair, including in models where healing is impaired. Mechanistically, the interest is in helping cells migrate into damaged areas and rebuild tissue, processes that extend beyond simply reducing pain or suppressing inflammation.
